1、電針對坐骨神經(jīng)損傷大鼠腦源性神經(jīng)營養(yǎng)因子的影響3800字 摘要:目的 觀察電針對坐骨神經(jīng)損傷(SNI)大鼠腦源性神經(jīng)營養(yǎng)因子(BDNF)的影響,探討其治療SNI的生物學(xué)機(jī)制。方法 選取成年雄性Wistar大鼠50只,分離并切斷大鼠坐骨神經(jīng)后于體式顯微鏡下將兩側(cè)斷端拉入神經(jīng)再生室內(nèi)造模。大鼠隨機(jī)分為正常組、假手術(shù)組、模型組和電針組。電針組進(jìn)行電針治療,連續(xù)28 d。治療結(jié)束后,HE染色觀察神經(jīng)再生,免疫熒光檢測神經(jīng)組織和脊髓BDNF的表達(dá),ELISA檢測血清BDNF的表達(dá)。結(jié)果 電針組軸突再生數(shù)量明顯多于模型組,且大體輪廓較清晰;與模型組比較,電針組可促進(jìn)大鼠神經(jīng)組織、脊髓和血清BDNF的表達(dá)(

Methods Fifty adult male Wistar rats were chosen, and the sciatic nerves of rats were cut off and pulled on both sides of the cut ends into nerve regeneration chamber. The rats were randomly divided into normal group, sham-oper
4、ation group, model group, and electroacupuncture group. In the electroacupuncture group, the rats were treated by electroacupuncture for 28 days. After the treatment, the nerve regeneration was observed through HE staining. Immunofluorescence was used to analyze the expression changes of BDNF in the
5、 nerve tissue and spinal cord. ELISA was used to observe the changes of expression of serum BDNF. Results The amount of axon regeneration in the electroacupuncture group was obviously more than that in the model group, and the outline of the tissue more clear. Electroacupuncture could promote the ex
6、pression of BDNF in the nerve, spinal cord and serum of SNI of rats compared with model group (P0.01). Conclusion Electroacupuncture can promote the repairment and regeneration of SNI in rats by upregulating the expression of BDNF.Key words: electroacupuncture; sciatic nerve; brain derived neurotrophic factor; axon; rats周?神經(jīng)損傷(PNI)屬中醫(yī)學(xué)“傷筋”“痿證”范疇。諸病源候論?金瘡傷筋斷骨候云:“夫金瘡始傷之時,半傷其筋,榮衛(wèi)不通,其瘡雖愈合后,仍令痹不仁也。”目前治療方法有神經(jīng)吻合技術(shù)、細(xì)胞移植及相關(guān)藥物應(yīng)用等,但神經(jīng)功能的恢復(fù)仍不理想1-2。實(shí)驗(yàn)和臨床研究表明,電針在損傷神經(jīng)的功能恢復(fù)方面療效肯定和優(yōu)勢明顯,能明顯提高PNI患肢的肌力,促進(jìn)神經(jīng)功能的恢復(fù),減輕后遺癥3-4?;痦?xiàng)目:國家自然科學(xué)基金(81373754)通訊
